Immunoprecipitation Various amounts of Rabbit anti MLH1 N-terminal antibody (M8320) were used to immunoprecipitate MLH1 from MCF-7 cell lysates. Detection antibody: Monoclonal antibody to MLH1. Lanes 1. 15 μg of antibody 2. 5 μg of antibody 3. 1 μg of antibody 4. Negative control – without cell lysate 5. Negative control – without IP antibody 6. Positive control – Total MCF-7 cell lysate direct labeling
Immunofluorescence MCF7 cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.2% Triton X-100. Fixed cells were stained with 5 μg/mL Anti-MLH1 (N-terminal) antibody produced in Rabbit (Cat. No. M8320) followed by 1:500 Anti-Rabbit IgG-Cy3 antibody. Picture B: cells were counterstained with DAPI (blue) to stain nuclei.

Biochem/physiol Actions: MutL protein (MLH), a homolog of the E. coli MutL gene, is involved in DNA mismatch repair. Nonpolyposis colorectal cancer-2 is caused by a hereditary mutation in the MLH1 gene. This cancer can also be a result of hypermethylation of one MLH1 allele in somatic cells (a germline epimutation).

General description: MutL homolog 1 (MLH1) is a nucleoprotein and a major component of mismatch repair system. The MLH1 gene is localized on chromosome 3p21 and is made up of 19 exons. The protein has a molecular weight of 80kDa.
General description: MutL homolog 1 (MLH1) is part of a large multi-subunit protein complex of tumor suppressors, DNA damage sensors and signal transducers, named BRCA1-associated genome surveillance complex (BASC).
Immunogen: synthetic peptide corresponding to amino acids 60-75 of human MLH1, conjugated to KLH via an N-terminal added cysteine residue. The corresponding peptide sequence is conserved in human, rat, and mouse.
Physical form: Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
