Immunoprecipitation Biotin conjugated SILu™Rich Anti-IL6 MAb, Cl: IL6-4G1 (Cat. No. MSAB001) was used to immunoprecipitate IL6 from concentrated supernatant of IL-1β activated MG-63 cells. Detection antibody: Rabbit Anti-IL6, followed by Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1.1 μg/test antibody 2. 2 μg/test antibody 3. Negative control: 1 μg/test non-relevant antibody 4. Concentrated supernatant of IL-1b activated MG-63 cells (~11 ng/lane).

ImmunoAffinity Enrichment-Mass Spectrometry (IAE-MS) Biotin conjugated SILu™Rich Anti-IL6 MAb, Clone:IL6-4G1 (Cat. No. MSAB001) was used to immunoprecipitate recombinant, stable isotope-labeled human IL-6 (SILu™Prot IL6) internal MS standard (Cat. No. MSST0017) and endogenous IL6 from concentrated supernatant of MG-63 cells treated with IL-1β. The enriched proteins were further digested and the tryptic pEFLQSSLR peptide was analyzed by Multiple Reaction Monitoring (MRM-MS) chromatograms of its y4 transition.
