Immunohistochemistry Staining of methacarn-fixed, formic acid-treated Alzheimer′s Disease (AD) brain sections, with 1:1,000 Rabbit Anti-Nitrotyrosine (Cat. No. N0409) using biotin/ExtrAvidin®-Peroxidase.
Immunofluorescence
A549 cells metabolically labeled with 3-nitro-L-tyrosine were fixed and permeabilized with methanol followed by aceton. Fixed cells were stained with 1:500 Rabbit Anti-Nitrotyrosine (Cat. No. N0409). The antibody was developed using Goat Anti-Rabbit IgG, FITC conjugate.
Immunofluorescence A549 cells metabolically labeled with 3-nitro-L-tyrosine were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 1:1,000 Rabbit Anti-Nitrotyrosine (Cat. No. N0409). The antibody was developed Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No. F9887). Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting Tissue lysates were separated on SDS-PAGE and probed with 1:1,000 Rabbit Anti-Nitrotyrosine (Cat. No. N0409). The antibody was developed using Goat Anti-Rabbit IgG-peroxidase (Cat. Biochem/physiol Actions: Anti-Nitrotyrosine recognizes nitrated proteins and 3-nitro-L-tyrosine. It does not cross-react with L-tyrosine, ρ-nitro-L-phenylalanine, 3-amino-L-tyrosine, 3-chloro-L-tyrosine and phospho-L-tyrosine BSA conjugates.
Biochem/physiol Actions: Peroxynitrite has been shown to promote the nitration of tyrosine residues in proteins, and oxidation of redox metal centers, DNA, lipids and cysteine and methionine residues in proteins, suggesting that it plays a major role in oxidative cellular damage. Peroxynitrite has been implicated in the pathogenesis of several inflammatory, infectious and degenerative human disease, including neurological conditions such as Alzheimer′s disease, amyotrophic lateral sclerosis (ALS), in the pathogenesis of asthma, atherosclerosis and a variety of conditions precipitated by endothelial injury.
Biochem/physiol Actions: Reacts with peroxynitrite-nitrated proteins and 3-nitro-L-tyrosine. Does not crossreact with L-tyrosine, p-nitro-L-phenylalanine, 3-amino-L-tyrosine, 3-chloro-L-tyrosine, and phospho-L-tyrosine. Binding of the antibody is blocked by 3-nitrotyrosine.

General description: Peroxinitrite (ONOO-), a potent oxidant linked to cellular oxidative damage, is an important member of the family of reactive oxygen and nitrogen species, rapidly generated in vivo from nitric oxide and superoxide anion. Nitrotyrosine is a product of tyrosine nitration, that is facilitated by reactive nitrogen species.
Immunogen: nitrated KLH (Keyhole Limpet Hemocyanin).
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
Preparation Note: Purified by affinity chromatography on a column of 3-nitro-L-tyrosine.
