| Biochem/physiol Actions: | Cancer/testis (CT) antigens are the protein products of genes that are activated in a wide variety of tumors and can elicit autologous cellular and humoral immune responses. Their normal expression is restricted to male germ cells in the testis and they are not expressed in adult somatic tissues. Therefore, CT antigens are promising candidates for cancer immunotherapy. Approximately 20 CT antigens or antigen families have been identified to date. NY-ESO-1 is a CT protein to which cell-mediated immune response has been demonstrated. It is highly expressed in various types of cancers. For example, patients with melanoma contain tumor-infiltrating lymphocytes cells (TILs) that recognized NY-ESO-1 protein. The expression of genes encoding CT antigens was examined in sporadic medullary thyroid carcinoma. The expression of NY-ESO-1 was found in 65 percent of the samples and correlated with tumor recurrence in primary MTC tumors. Furthermore, high frequency of expression was found in breast cancer especially in benign lesions and in multiple myeloma. Due to its wide expression in different cancers, NY-ESO-1 protein was used as a vaccine in cancer patients. |
