Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-NPTII antibody produced in Rabbit (Cat. No. N6537) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-transgenic Nicotiana tabacum leaves over expressing E. coli NPTII (Lane 1) while no staining is detected in the wild type sample-Nicotiana tabacum leaves (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate.
Western Blotting Whole extract (cytosolic fraction) from the leaves of transgenic Nicotiana tabacum expressing E. coli NPTII were separated on SDS-PAGE and probed with Anti-NPTII antibody produced in Rabbit (Cat. No. N6537). The antibody was developed using 1:10,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Application: Anti-NPTII antibody has been used in immunoblotting.
Biochem/physiol Actions: The antibody recognizes bacterial NPTII expressed in transgenic tobacco plants.
Biochem/physiol Actions: The Neomycin phosphotransferase II (NPTII) gene confers resistance to some aminoglycoside antibiotics including neomycin and kanamycin in bacteria and plant cells and to G418 (Gibco Geneticin reagent) sulfate in mammalian cells. NPTII phosphorylates and inactivates kanamycin, preventing kanamycin from binding to the 30S ribosomal subunit to inhibit protein synthesis, thus rendering cells resistant to the antibiotic. NPTII serves as a marker for the selection of genetically engineered crops.

General description: Neomycin phosphotransferase II (NPTII) gene also known as the kanamycin-resistance marker gene, encoding for NPTII (30 kDa) an aminoglycoside 3′ -phosphotransferase II (APHII), was isolated from the E. coli transposon Tn5.
Immunogen: synthetic peptide corresponding to amino acids 133-150 located at the mid-region of E. coli NPTII, conjugated to KLH.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
