Immunofluorescence Cytoplasmic and nuclear staining of NR4A2 in NIH-3T3 cells. Fixed cells were stained with 3 μg/mL Rabbit Anti-NR4A2 (Cat. No. N6538) followed by Goat Anti-Rabbit IgG, FITC conjugate (x40 magnification).
Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-NR4A2 antibody produced in Rabbit (Cat. No. N6538) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing NR4A2 (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1.5 μg/mL.
Immunoblotting Cell lysates of HEK-293T overexpressing human NR4A2 (Lanes 1,3) and non-transfected (Lane 2) were separated on SDS-PAGE, blotted with Anti-NR4A2 (Cat. No. N6538) and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and chemiluminescent substrate. Lanes 1. Antibody 1.5 μg/mL 2. Antibody 1.5 μg/mL 3. Antibody 1.5 μg/mL + NR4A2 immunizing peptide (human, 160-179)
Immunoblotting Cell lysates were separated on SDS-PAGE, blotted with Anti-NR4A2 (Cat. No. N6538) and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and chemiluminescent substrate. Application: Anti-NR4A2 antibody produced in rabbit has also been used in immunohistochemistry (IHC).
Application: Anti-NR4A2 antibody produced in rabbit has been used in western blotting.
Biochem/physiol Actions: Anti-NR4A2 specifically recognizes human NR4A2.
Biochem/physiol Actions: Nuclear receptor subfamily 4 group A member 2 (NR4A2) acts as an activator of tyrosine hydroxylase. It is crucial for the process of DNA repair by melanocortin-1 receptor.
Biochem/physiol Actions: Nuclear receptor subfamily 4 group A member 2 (NR4A2) receptors function as transcription factors. NR4A2 binds the NGFI-B (nerve growth factor-induced clone B) response element (NBRE) sequence (AAAGGTCA) in the form of monomer. NR4A2 is induced by multiple extracellular signals, including fatty acids, stress, prostaglandins, growth factors, calcium, inflammatory cytokines, peptide hormones, and neurotransmitters in a cell type specific manner. Cyclic adenosine monophosphate (cAMP) triggers the hepatic expression of NR4A2 in response to glucagon and fasting. Diabetic mice with elevated gluconeogenesis also show high levels of NR4A2. NR4A2 is essential for the differentiation of nigral dopaminergic neurons. Mice lacking Nurr1 failed to generate midbrain dopaminergic neurons. NR4A2 mutations have been associated with disorders related to dopaminergic dysfunction, including Parkinson′s disease, schizophrenia and manic depression.

General description: NR4A2 is encoded by the gene mapped to human chromosome 2q24.1.
General description: Nuclear receptor subfamily 4 group A member 2 (NR4A2) is part of transcriptional regulator subfamily, under the family of nuclear hormone receptors. It is a transcription factor expressed in the brain and T-cells.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note: For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in “frost-free” freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
