Human Jurkat cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 5 μg/mL Anti-ORAI1 antibody produced in Rabbit (Cat. No. O8264). The antibody was developed with Goat Anti-Rabbit IgG, Cy3™ conjugate. Pictures B, C: cells were counterstained with DAPI (blue) to stain nuclei.
Western Blotting Whole extract of Human HEK-293T cells expressing ORAI1 was separated on SDS-PAGE and probed with Anti-ORAI1 antibody produced in Rabbit (Cat. No. O8264). The antibody was developed using 1:30,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Lanes 1. 0.5 μg/mL antibody 2. 1 μg/mL antibody 3. 1 μg/mL antibody + inhibition with immunizing peptide 4. 2 μg/mL antibody 5. Negative control

Application: Anti-ORAI1 antibody produced in rabbit has been used in immunohistochemistry and Western blotting.
Biochem/physiol Actions: ORAI calcium release-activated calcium modulator 1 (ORAI1) modulates Ca2+ entry into cells. The channel is activated by stromal interaction molecule-1 (STIM-1). Mutation in the gene encoding the protein has been linked to late-onset tubular aggregate myopathy and congenital miosis.

General description: ORAI calcium release-activated calcium modulator 1 (ORAI1) is the pore-forming subunit of the Ca2+ release-activated (CRAC) channel. The protein possesses four predicted transmembrane domains and intracellular amino and carboxyl-terminal domains. The ORAI1 gene is localized on human chromosome 12q24.31.
Immunogen: synthetic peptide corresponding to amino acids 288-301 of human ORAI1, conjugated to KLH. The corresponding sequence differs by 2 amino acids in rat and mouse.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
