Application: Kinase activity is measured as the molar amount of phosphate incorporated into the CREBtide substrate peptide per minute per mg protein at 30 °C using a final concentration of 50 μM [32P] ATP.
Biochem/physiol Actions: A catalytic subunit of cAMP-dependent protein kinase, the protein encoded by PRKACB catalyzes events downstream of GPCRs including cell cycle, differentiation and proliferation. When activated, this subunit acts on metabolic enzymes, ion channels and transcription factors such as CREB.
Biochem/physiol Actions: cAMP-dependent protein kinase (PKA; EC 2.7.1.37) is an essential enzyme in the signaling pathway of the second messenger cAMP. Through phosphorylation of target proteins, PKA controls many biochemical events in the cell including regulation of metabolism, ion transport, and gene transcription.
General description: Protein kinase cAMP-activated catalytic subunit β (PRKACB) is one of the catalytic subunits of the serine/threonine kinase, protein kinase A. The gene encoding it is localized on human chromosome 1p36.1.

The molecular weight of about 65kDa is confirmed by the SDS-PAGE gel. The product was generated by expressing recombinant full-length human PKAcβ2 with an additional N-terminal GST tag by baculovirus in Sf9 insect cells. The gene accession number is NM_002731. The resulting protein is 351aa cAMP-dependent protein kinase catalytic subunit beta isoform 2 (NP_002722), or according to UniProt, Isoform 1 (P22694-1).
Other Notes: One unit will incorporate one nanomole of phosphate into the Crebtide (KRREILSRRPSYR) substrate per minute at 30 deg C and pH 7.2 using a final concentration of 50 μM [32P] ATP.
Physical form: Supplied as a solution of 5 μg in 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 0.25 mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF and 25% glycerol.
