Western Blotting Rabbit Anti-PCNA Antibody, Affinity Purified: Cat. No. PLA0079: Detection of Human PCNA by Western Blot and Immunoprecipitaiton. Samples: A. Whole cell lysate (WCL) from A-431 cells. B. WCL (500 μg for IP; 30 μg for WCL lane) from MDA-MB-468 cells. Antibodies: A. Affinity purified Rabbit Anti-PCNA Antibody (Cat. No. PLA0079) used at the indicated concentrations for WB. B. PCNA was immunoprecipitated using Antibodies recognizing different epitopes (including Cat. No. PLA0080) or a Mouse monoclonal Antibody to PCNA (mAb). Control mock IP was performed using normal Mouse IgG (M-IgG) and normal Rabbit IgG (R-IgG). Immunoprecipitated PCNA was blotted using one of the alternate Antibodies (each at 1 μg/mL). Detection: Chemiluminescence with an exposure time of 2 minutes (A) or 5 seconds (B).
Flow Cytometry Rabbit Anti-PCNA Antibody, Affinity Purified: Cat. No. PLA0079: Flow Cytometric Detection of PCNA Versus DNA Content. Asynchronous Jurkat cells were fixed and permeabilized in a sequential treatment of FACS buffer (PBS, 0.5% Triton-X-100, 0.5 mM EDTA, 1% BSA) and 100% methanol. 1 × 106 cells were stained with 0.03 μg Anti-PCNA (PLA0079). Secondary detection was performed with FITC conjugated Goat F(ab′)2 Anti-Rabbit Antibody, and DNA stained with PI.

| Biochem/physiol Actions: | Proliferating cell nuclear antigen (PCNA) protein functions as an auxiliary factor of polymerase δ. It acts as a cofactor to polymerase ε, polymerase β, and translesion synthesis polymerases, such as η, κ, λ, and θ. In chronic lymphoid leukemia (CLL) patients, intracellular levels of PCNA protein can be used as a marker to identify clinical behavior and survival. |

| General description: | Proliferating cell nuclear antigen (PCNA), a single-copy gene, spans about 12 kb. This gene is located on human chromosome 20p13. PCNA is expressed at high levels in the thymus, bone marrow, fetal liver, and some cells of the small intestine and colon. It is located in the nucleus. |
| Immunogen: | The epitope recognized by PLA0079 maps to a region between residues 75 and 125 of human proliferating cell nuclear antigen using the numbering given in SwissProt entry P12004 (GeneID 5111). |
| Other Notes: | PCNA (Proliferating Cell Nuclear Antigen) was first identified as a factor expressed in the nuclei of replicating cells. PCNA plays a role in both DNA synthesis and repair. PCNA functions as a processivity factor for DNA polymerase by acting as a DNA clamp that tethers the polymerase catalytic units to the DNA template. The structure of PCNA is that of a trimeric ring that encircles and slides along replicating DNA. PCNA interacts with and is regulated by several proteins involved in cell cycle control and check point processes. |
