Western Blotting Rabbit Anti-MAD1 Antibody, Affinity Purified: Cat. No. PLA0092: Detection of Human MAD1 by Western Blot and Immunoprecipitation. Samples: Whole cell lysate (50 μg/lane in A; 50 μg input in B, 1 mg for IP in B) from HeLa cells. In A, HeLa cells were treated with Nocodazole (+) or mock treated (-). Antibody: Affinity purified Rabbit Anti-MAD1 Antibody (Cat. No. PLA0092) used at 0.1 μg/mL for Western Blot (A) and at 1 μg/mg lysate for IP (B). MAD1 was also immunoprecipitated using Rabbit Anti-MAD1 Antibodies recognizing different epitopes using 1 μg/mg lysate. In B, MAD1 in input and immunoprecipitate was detected using Antibody at 0.2 μg/mL. Detection: Chemiluminescence with exposure times of 10 minutes (A) and 5 minutes (B).

Other Notes: MAD1 (Mitotic Arrest Deficiency 1) is a component of the mitotic spindle-assembly checkpoint that ensures that all chromosomes are aligned at the metaphase plate before the onset of anaphase. MAD1 functions as a homodimer that interacts with and regulates MAD2. MAD1 may play a role in tumor suppression.
