Samples: NBF-fixed, asynchronous HeLa cells. Antibody: Affinity purified Rabbit Anti-Phospho KAP-1 (S824) (Cat. No. PLA0140) used at a dilution of 1:200 (1 μg/mL). Detection: Red fluorescent Anti-Rabbit IgG - DyLight® 594 conjugated used at a dilution of 1:100.
Western Blotting Rabbit Anti-Phospho KAP-1 (S824) Antibody, Affinity Purified: Cat. No. PLA0140: Detection of Human Phospho KAP-1 (S824) by Western Blot of WCL and Immunoprecipitates. Samples: Whole cell lysate (50 μg for WCL lanes; 1 mg for IP, 20% of IP loaded) from asynchronous HeLa cells that were either mock treated or treated with neocarzinostatin (NCS; 200 ng/mL, 30 minutes). Antibodies: Affinity purified Rabbit Anti-phospho KAP-1 (S824) Antibody PLA0140 used for WB at 0.1 μg/mL. KAP-1 was immunoprecipitated using Rabbit Anti-KAP-1 Antibody PLA0078 at 6 μg/mg lysate. To examine total KAP-1, the blot was stripped and then blotted with PLA0078 at 1 μg/mL. Detection: Chemiluminescence with exposure times of 30 seconds (upper panel) and 1 second (lower panel).

Samples: Whole cell lysate (1 mg for IP; 20% of IP loaded) from NIH3T3 cells that were mock treated (-) or treated with Etoposide (100 μM, 2h). Antibodies: Affinity purified Rabbit Anti-Phospho KAP-1 (S824) Antibody PLA0140 used for IP at 6 μg/mg lysate. Phospho KAP-1 (S824) was also immunoprecipitated by Rabbit Anti-KAP-1 Antibody PLA0078. For blotting immunoprecipitated Phospho KAP-1 (S824), PLA0140 was used at 0.2 μg/mL. Detection: Chemiluminescence with an exposure time of 3 minutes.

KAP-1 also appears to function as a scaffold for chromatin-remodeling complexes involved in transcriptional repression. |
