Immunoprecipitation Rabbit anti-STAT3 Antibody, Affinity Purified: Cat. No. PLA0255: Detection of Human STAT3 by Western Blot of Immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified Rabbit Anti-STAT3 Antibody Cat. No. PLA0255 used for IP at 3 μg/mg lysate. STAT3 was also immunoprecipitated by Rabbit Anti-STAT3 Antibody B, which recognizes a downstream epitope. For blotting immunoprecipitated STAT3 it was at 0.4 μg/mL. Detection: Chemiluminescence with an exposure time of 10 seconds.

Immunogen: The epitope recognized by PLA0255 maps to a region between residue 675 and 725 of human signal transducer and activator of transcription 3 using the numbering given in entry NP_644805.1 (GeneID 6774).
Other Notes: STAT3 (signal transducer and activator of transcription 3) is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated through phosphorylation in response to various cytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. This protein mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis. The small GTPase Rac1 has been shown to bind and regulate the activity of this protein. PIAS3 protein is a specific inhibitor of this protein [taken from NCBI Entrez Gene (GeneID:6774)].
