Biochem/physiol Actions: TMI-1 (WAY-171318) is potent inhibitor of disintegrin metalloenzyme 17 (ADAM17) also known as the tumor necrosis factor-α (TNF-α) converting enzyme (TACE), and matrix metalloprotease (MMP). TMI-1 potently inhibits LPS-induced TNF-α secretion in human primary monocytes, and human whole blood. TMI-1 selectively induces caspase-dependent apoptosis in triple negative (TN) and ERBB2-overexpressing breast tumor cell lines.
