A. A431 cell extract (Human Epidermoid carcinoma) was separated by SDS-PAGE and probed with (Lane 1) of without (Lane 2) Monoclonal Anti-RAF-1 (Clone RNP1, Cat. No. R2404) followed by Goat Anti-Mouse, Peroxidase conjugate (Cat. No. A2304, Lane 2). B. Localization of RAF-1 protein in the cytoplasm of A431. A431 cells were fixed with 4% paraformaldehyde and permeabilized with 0.2% Triton X-100. The cells were incubated with Monoclonal Anti-RAF-1 (Clone RNP1, Cat. No. R2404) and further developed with Rabbit Anti-Mouse IgG, FITC-conjugate.

Biochem/physiol Actions: Monoclonal anti-Raf-1/c-Raf antibody recognizes human, rat and mouse Raf-1 (~75 kDa). The product may also detect Raf-1 degradation products as additional weaker bands at approx. 50 kDa.
Biochem/physiol Actions: Raf-1 (rapidly accelerated fibrosarcoma) controls cell growth and differentiation. Its oncogenic form can initiate malignant transformation of cells. Raf-1 has a critical role in the Ras/MAP kinase signalling pathway, integrating mitogenic signals in mammalian cells from many growth factors, cytokines and oncogenes, e.g., epidermal growth factor (EGF), platelet derived growth factor (PDGF), insulin, interleukin-2 (IL-2) and IL-3, granulocyte macrophage colony-stimulating factor (GM-CSF) and CSF-1. The regulation of Raf-1 activity is a highly complex and involves multistep process. Inactive Raf-1 is found in the cytosol, and is constitutively associated with the chaperones heat shock protein 90 (Hsp90), p50, and the 14-3-3 protein. Raf-1 activity is regulated by autophosphorylation at threonine residues and by phosphorylation at multiple serine and tyrosine residues by a number of protein kinases, including protein kinase C (PKC), non-receptor tyrosine kinase (Src) and Janus kinase 2 (JAK2).

General description: Monoclonal Anti-Raf-1/c-Raf (mouse IgG1 isotype) is derived from the RNP1 hybridoma produced by the fusion of mouse myeloma cells (NS1) and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to amino acids 334-345 of human Raf-1.
General description: Raf-1 is a serine/threonine kinase that is activated by tyrosine kinases and functions upstream of the MAPK-signaling pathway. Raf-1 inhibits Ras signaling and subsequently modulates cell proliferation and development. c-Raf expression has been linked to the pathogenesis of lung carcinoma.
Immunogen: Synthetic peptide corresponding to amino acids of human Raf-1.
Packaging: 200 μL in glass bottle
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note: For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
