| Biochem/physiol Actions: | Vascular endothelial growth factor (VEGF) is a critical factor in regulating angiogenesis in both physiological and pathological conditions. Transforming growth factor-α (TGF-α), TGF-β and platelet derived growth factor can potentially activate VEGF activity. VEGF mediates its activity through tyrosine kinase receptors, vascular endothelial growth factor receptor-1 (VEGFR-1) and VEGFR-2. VEGF produced by tumour cells and macrophages, plays a key role in cancer development by promoting endothelial cells proliferation and increasing vascular permeability. Enhanced expression of VEGF plays a key role in urologic neoplasm like bladder cancer, renal cell carcinoma, and cervical cancer. Polymorphism in VEGF gene increases the disease susceptibility. Blocking of VEGF mediated signalling pathway is a potential therapeutic strategy for cancer patients. |
