Streptolysin O is an antigenic protein capable of forming pore complexes in mammalian cells. The monomer molecular weight is reported to be 64–69 kDa. Pore complexes are reported to be up to 30 nm in diameter and appear to be comprised of a double wall containing as many as 125 monomers.
Hemolysis activity assay: Streptolysin O from Streptococcus pyogenes (Cat. No. S0149) was serially diluted (2-fold, top to bottom of plate) and incubated with erythrocytes (RCB) to calculate minimum Hemolysis titer.

Application: Permeabilizes membranes to permit cellular uptake of large or charged molecules.
Application: Streptolysin O from Streptococcus pyogenes has been used in streptolysin O (SLO) permeabilization. It has also been used to evaluate the barrier integrity and cytotoxicity of caco-2 and T84 IEC polarized monolayers.
Biochem/physiol Actions: Streptolysin O (SLO), obtained from Streptococcus pyogenes has the capability to block osteoclastogenesis, induced by receptor activator of nuclear factor κ-Β ligand (RANK) through the NF-κB signaling pathway. Streptococcus pyogenes (GAS) is a bacterial strain that results in bacterial arthritis, osteomyelitis and implant infections.
Biochem/physiol Actions: Thiol-activated toxin that permeabilizes animal cell membranes. The protein binds as a monomer to membrane cholesterol and subsequently polymerizes into large arc- and ring-shaped structures surrounding pores of >12 nm.

Other Notes: One unit will cause 50% lysis of 2% red blood cell suspensions in phosphate buffered saline, pH 7.4, after incubation at 37 °C for 30 min.
