Application: Anti- S6 kinase antibody may be used for detection by immunoblotting at a working dilution of 1:10,000 using mouse NIH 3T3 fibroblast cell extract. Detection by immunoblotting was also done in primary cultures of mouse glial cells and in LnCAP, prostate cancer cell line.
Biochem/physiol Actions: The sequence of the immunizing peptide is identical in rat and mouse S6K.

General description: S6 kinases are Ser/Thr protein kinases that are capable of phosphorylating the 40S ribosomal protein S6 in response to mitogen stimulation. S6 kinase (p70s6k) is activated by an upstream kinase, mTOR in response to nutrients and growth factors. S6 kinase (p70s6k) is a critical regulatory component and mTOR along with the eukaryotic initiation factor 4E (eIF4E)–binding protein 1 (4E-BP1) and S6 kinase (p70s6k) leads to increased ribosomal biogenesis and hence translation. As a critical component and has the ability to modify the cellular responses to availability of nutrients, and growth factors like insulin and IGF-1 when mTOR is chronically active. In presence of excess and chronic insulin, or high fat diets, S6 kinase (p70s6k) regulates by a negative feedback loop and constitutively shuts down the responsiveness of cells to insulin. Thus mTOR through the S6 kinase (p70s6k) plays a pivotal role in the development of insulin resistance and type 2 diabetes. mTOR signaling via p70S6 kinase also drives cell growth and proliferation, cancer and aging.
Anti-S6 Kinase reacts specifically with S6 kinase (70 kDa) in human, rat and mouse.
Immunogen: synthetic peptide corresponding to amino acids 6-23 of human S6 kinase.
