Enhanced Validation-By Independent Antibodies Immunofluoresence. NRK cells were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 10 μg/mL Anti-Superoxide Dismutase (MnSOD) (DD-17) antibody produced in Rabbit (Cat. No. S5069) (A). The antibody developed using Goat Anti-Rabbit IgG, Cy3 Conjugate antibody, and 1:500 Anti-Superoxide Dismutase (MnSOD) (KC-19) antibody produced in Rabbit (Cat. No. S5569) (B). The antibody was developed using Goat Anti-Rabbit IgG, Cy3 Conjugate antibody. Results Two Anti Superoxide Dismutase (MnSOD) antibodies, S5069 (A) and S5569 (B) target different regions of Superoxide Dismutase (MnSOD) show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunohistochemistry Formalin fixed, paraffin-embedded Human heart tissue sections were stained with 10 μg/mL Anti-Superoxide Dismutase (MnSOD) (DD-17) antibody produced in Rabbit (Cat. No. S5069), followed by 20 μg/mL Anti-Rabbit IgG (whole molecule)-Biotin antibody produced in Goat (Cat. No. B8895) and by 20 μg/mL ExtrAvidin®-Peroxidase (Cat. No. E2886).
Immunoblotting
Cell extract of human HeLa mitochondria was separated on SDS-PAGE and blotted with 1 μg/mL Rabbit Anti-Superoxide Dismutase (MnSOD) (DD-17) (Cat. No. S5069) and developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate.

Application: The antibody may be detected by various immunochemical techniques including immunoblotting, co-immunoprecipitation assay (co-IP), immunoprecipitation, indirect immunofluorescence and immunohistochemistry.
Biochem/physiol Actions: Superoxide Dismutase (SOD) catalyzes the dismutation of the cytotoxic superoxide radical (O2 -) to molecular oxygen and hydrogen peroxide. Manganese SOD (MnSOD) has been proposed to function as a tumor suppressor by modulating the activity of redox-sensitive transcription factors and specific signal mediators. MnSOD is involved in detoxification.

General description: Superoxide Dismutase (SOD) is a metalloprotein enzyme. It is widely distributed in the animal and plant kingdoms. The three common mammalian endogenous isozymes are the homodimeric copper-zinc SOD (Cu,ZnSOD, SOD1) found primarily in the cytosol, the homotetrameric glycosylated copper-zinc SOD, extracellular SOD (EC-SOD, SOD3), and the homotetrameric mitochondrial manganese SOD (MnSOD, SOD2). All three are nuclear-encoded. MnSOD is composed of four subunits, each containing one Mn2+.
Immunogen: synthetic peptide corresponding to amino acid residues 183-199 of human superoxide dismutase (MnSOD) with N-terminal added cysteine, conjugated to KLH. The corresponding sequence is conserved in many eukaryotes.
