Immunofluorescence 293-T expressing human E-Selectin cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.2% Triton X-100. Fixed cells were stained with 2 μg/mL Anti-E-Selectin (CD62E) antibody, Mouse monoclonal, Clone: 12B6 (Cat. No. S9555) followed by 1:500 Anti-Mouse IgG (whole molecule) F(ab′)2 fragment-Cy3™ antibody produced in Sheep (Cat No. C2181).

Biochem/physiol Actions: E-selectin acts as a key inflammatory cytokine. E-selectin-dependent rolling helps in binding pancreatic cancer cell to hyaluronic acid. In addition, the encoded protein in association with P-selectins, plays a vital role in recruiting leukocytes to inflammatory sites. Mutation in the gene is associated with the development of various diseases such as atherosclerosis and stroke.
Biochem/physiol Actions: Reacts specifically with human CD62E (E-Selectin, ELAM-1) expressed on the surface of activated endothelial cells. The antibody may weakly cross-react with recombinant hman P-Selectin and human platelet P-Selectin. Cross-reactivity has been observed with porcine E-Selectin. Monoclonal Anti-human E-Selctin inhibits adhesion of neutrophils, eosinophils, and a skin-homing T-cell subpopulation to activated endothelial cells and CD62E-transfected cells. 5th Workshop: code no. Biochem/physiol Actions: Selectins play a vital role in maintaining homoeostasis and cutaneous inflammation. SEL-E and SEL-P help in carrying T helper (Th1) cells into inflamed tissues. SEL-E acts as a receptor for cluster of differentiation 44 (CD44 (HCELL)), Mac-2 binding protein, mucin 1 (Muc-1), death receptor-3 and E-selectin ligand-1 (ESL-1). Overexpression of the gene might affect adhesion and migration of the cells leading to the endometriosis disorder.

General description: CD62E (E-Selectin) surface antigen is a glycoprotein and member of the selectin family of adhesion receptors. It is expressed by activated human endothelial cells and is detectable in the plasma.
General description: Monoclonal anti-E-selectin (CD62E) (mouse IgG1 isotype) is derived from the 1.2B6 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with TNF-α activated human endothelial cells. This antibody reacts specifically with E-selectin (CD62E) expressed on the surface of activated endothelial cells. The antibody may weakly cross-react with recombinant human P-selectin and human platelet P-selectin. Cross-reactivity has been observed with porcine E-selectin.
It is a 140KDa protein mainly expressed in endothelial cells. E- selectin is characterized with an N-terminal domain, epidermal growth factor (EGF)-like domain, lectin (ligand-binding) domain, variable short consensus repeats and transmembrane domain.
General description: Selectin E (SELE) is encoded by the gene mapped to human chromosome 1q24.2. The encoded protein is mainly expressed on luminal surfaces of inflamed vasculature.
Immunogen: TNF-α activated human endothelial cells.
Physical form: Solution, 0.2 μm filtered, in 0.01 M phosphate buffered saline, pH 7.4.
Preparation Note: For continuous use, store sterile at 2-8°C. For extended storage freeze in sterile working aliquots. Repeated freezing and thawing, or storage in "frostfree" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
