| Biochem/physiol Actions: | TGFBR1 (transforming growth factor β receptor 1) stimulates signaling pathway that mediates cell growth and division. The complex formed by TGFBR2 with TGFBR1 results in the phosphorylation and activation of TGFBR1. TGFBR1 is responsible for the activation of canonical Smad (mothers against decapentaplegic homolog 3) proteins and transcriptional factors Snail (zinc finger protein) and Twist (twist family bHLH transcription factor 1). The activation of a number of non-canonical effectors such as RhoA (ras homolog family member A), TAK1 (transforming growth factor-β-activated kinase 1) and Akt (RAC-α serine/threonine-protein kinase) is also associated with TGFBR1. The arrangement of the TGFBR1 receptor along with TGFBR2 at the focal adhesions mediates the multimerization and stimulation of TGFβ ligand. Downregulation of TGFBR1 is observed in breast cancer. TGFBR1 is also known to be associated with colorectal cancer. |
