Immunohistochemistry Immunohistochemical analysis of paraffin-embedded Cal27 xenograft, using NRF2 (SAB2701989) antibody at 1:500 dilution.
Immunofluorescence NRF2 antibody [N2C2], Internal detects NRF2 protein at nucleus by immunofluorescent analysis. Sample: HeLa cells were fixed in 4% paraformaldehyde at RT for 15 min. Green: NRF2 protein stained by NRF2 antibody [N2C2], Internal (SAB2701989) diluted at 1:500. Red: Phalloidin, a cytoskeleton marker, diluted at 1:200. Scale bar = 10 μm.

Western Blotting NRF2 antibody detects NRF2 protein by western blot analysis. Non-transfected (–) and NRF2-transfected (+, including 3xFlag-tag) 293T whole cell extracts (30 μg) were separated by 5% SDS-PAGE, and the membrane was blotted with NRF2 antibody (SAB2701989) diluted by 1:1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
Western Blotting Whole cell extract (30 μg) was separated by 5% SDS-PAGE, and the membrane was blotted with NRF2 antibody [N2C2], Internal (SAB2701989) diluted at 1:500. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
Western Blotting NRF2 antibody detects NRF2 protein by Western blot analysis. Un-treated (*#8211;) and treated (+, 30 μM tBHQ treatment for 4 hr) MDA-MB-231 nuclear extract (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with NRF2 antibody (SAB2701989) diluted at 1:500.
Immunoprecipitation Immunoprecipitation of NRF2 protein from HepG2 whole cell extracts using 5 μg of NRF2 antibody [N2C2], Internal (SAB2701989). Western blot analysis was performed using NRF2 antibody [N2C2], Internal (SAB2701989). EasyBlot anti-Rabbit IgG was used as a secondary reagent.
ChIP Assay ChIP was performed with HepG2 chromatin extract and 5 μg of either normal rabbit IgG or anti-NRF2 antibody. The precipitated DNA was detected by PCR with primer set targeting to GCLC gene locus.

Application: Suggested starting dilutions are as follows: ICC/IF: 1:100-1:1000, IHC-P: 1:100-1:1000, IP: 1:100-1:500, WB: 1:500-1:3000, ChIP assay: Assay-dependent dilution. Not yet tested in other applications. Optimal working dilutions should be determined experimentally by the end user.
Biochem/physiol Actions: NFE2 (MIM 601490), NFE2L1 (MIM 163260), and NFE2L2 comprise a family of human genes encoding basic leucine zipper (bZIP) transcription factors. They share highly conserved regions that are distinct from other bZIP families, such as JUN (MIM 165160) and FOS (MIM 164810), although remaining regions have diverged considerably from each other (Chan et al., 1995 [PubMed 7868116]).[supplied by OMIM]
The nuclear factor erythroid 2-related factor 2 (Nrf2) controls phase II detoxifying enzymes, antioxidants and efflux transporters like P-glycoprotein (P-gp). De novo mutations in NFE2L2 results in a multisystem disorder. It modulates the expression of more than 200 genes.

General description: The nuclear factor erythroid 2-related factor 2 (NFE2L2) is a major transcriptional factor. NRF2 gene codes for NFE2L2. It is a member of the cap ‘n’ collar (Cnc) family of basic leucine zipper transcription factors. It is located on human chromosome 2q31.
Immunogen: Recombinant fragment corresponding to a region within amino acids 108 and 413 of NRF2 (Uniprot ID#Q16236)
Physical form: 1XPBS, 20% Glycerol (pH7). 0.025% ProClin 300 was added as a preservative.
