Immunohistochemistry Brn2 antibody detects Brn2 protein at nucleus in mouse brain by immunohistochemical analysis. Sample: Paraffin-embedded mouse brain. Brn2 antibody (SAB2702086) diluted at 1:500.
Immunofluorescence Brn2 antibody detects Brn2 protein at nucleus by immunofluorescent analysis. Sample: SK-N-AS cells were fixed in 4% paraformaldehyde at RT for 15 min. Green: Brn2 protein stained by Brn2 antibody (SAB2702086) diluted at 1:1000. Red: beta Tubulin 3/ Tuj1, a cytoskeleton marker, stained by beta Tubulin 3/ Tuj1 antibody . Scale bar = 10 μm.
Western Blotting Whole cell extract (30 μg) was separated by 10% SDS-PAGE, and the membrane was blotted with Brn2 antibody (SAB2702086) diluted at 1:1000.
Immunoprecipitation Brn2 antibody immunoprecipitates Brn2 protein in IP experiments. IP Sample: 1000 μg 293T whole cell lysate/extract A. 50 μg 293T whole cell lysate/extract B. Control with 2 μg of preimmune rabbit IgG C. Immunoprecipitation of Brn2 protein by 2 μg of Brn2 antibody (SAB2702086) 10% SDS-PAGE The immunoprecipitated Brn2 protein was detected by Brn2 antibody (SAB2702086) diluted at 1:1000. EasyBlot anti-Rabbit IgG was used as a secondary reagent.
ChIP Assay Brn2 antibody immunoprecipitates Brn2 protein-DNA in ChIP experiments. ChIP Sample: 293T whole cell lysate/extract A. 5 μg preimmune rabbit IgG B. 5 μg of Brn2 antibody (SAB2702086) The precipitated DNA was detected by PCR with primer set targeting to MITF promoter.

Biochem/physiol Actions: POU3F2 belongs to a large family of transcription factors that bind to the octameric DNA sequence ATGCAAAT. Most of these proteins share a highly homologous region, referred to as the POU domain, that occurs in several mammalian transcription factors, including the octamer-binding proteins Oct1 (POU2F1; MIM 164175) and Oct2 (POU2F2; MIM 164176) and the pituitary protein Pit1 (PIT1; MIM 173110). Class III POU genes are expressed predominantly in the central nervous system (CNS). It is likely that CNS-specific transcription factors such as these play an important role in mammalian neurogenesis by regulating their diverse patterns of gene expression (Schreiber et al., 1993 [PubMed 8441633]; Atanasoski et al., 1995 [PubMed 7601453]).[supplied by OMIM]
