Immunofluorescence Cytoplasmic and nuclear localization of VAC14 in HeLa cells. Fixed cells were stained with 8 μg/mL Anti-VAC14 (Cat. No. SAB4200074) followed by Goat Anti-Rabbit IgG, Cy3 conjugate (×40 magnification). Nuclear staining with DAPI.
Immunoprecipitation Anti-VAC14 (Cat. No. SAB4200074), was used to immunoprecipitate human VAC14 from A431 cell lysate. Detection antibody: Anti-VAC14 (C-terminal) (Cat. No. SAB4200075). Lanes 1. Biochem/physiol Actions: VAC14 is a regulator of phosphoinositide kinase, FYVE-type zinc finger containing (PIP5K3/PIKfyve), a dual specificity kinase that phosphorylates phosphatidylinositol 3-phosphate (PtdIns3P). This generates housekeeping phospholipid PtdIns(3,5)P2, that controls multivesicular body morphology, retrograde traffic to the trans-Golgi network and is critical for neuronal survival. VAC14/ArPIKfyve upregulates PIKfyve phosphoinositide-5-kinase activity. In 3T3-L1 adipocytes, VAC14 and PIP5K3 interaction has been shown to play a critical role in insulin-regulated glucose transporter (GLUT4) translocation from intracellular storage compartment to the cell surface. Knockout of the VAC14 gene in mouse results in massive neurodegeneration and affects neurons in the midbrain and of the peripheral sensory system.

General description: VAC14 (VAC14 component of PIKFYVE complex) gene (also known as ArPIKfyve, TAX1BP2, TRX) is mapped within human chromosome 16q22.1-q22.2 and encodes a scaffold protein. This protein is a component of phosphoinositide kinase, FYVE-type zinc finger containing (PIKfyve) protein kinase complex.
