Immunofluorescence HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton™ X-100. Fixed cells were stained with 5μg/mL Rabbit Anti-Rab32 (Cat. No. SAB4200086). The antibody was developed using Goat Anti-Rabbit IgG, Atto 488 conjugate (Cat. No. 18772) (×100 magnification).
Immunoblotting Whole extract of human platelets was separated on SDS-PAGE and probed with Rabbit Anti-Rab32 (Cat. No. SAB4200086). The antibody was developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. 1 μg/mL antibody 2. 1 μg/mL antibody + Rab32 immunizing peptide (human, 201-214) 3. 2 μg/mL antibody

Application: Anti-Rab32 antibody produced in rabbit has been used for western blotting and immunofluorescence.
Biochem/physiol Actions: RAB32 (Ras-related protein) controls intracellular lipid accumulation in hepatocytes. Absence of it promotes the expression of ATGL (adipose triglyceride lipase) and thereby causes lipolysis. RAB32 is involved in hepatic steatosis. In melanogenesis, it is involved in the transport of important enzymes, such as tyrosinase and Tyrp1 (tyrosinase related protein 1). RAB32 also interacts with and is involved in localization of leucine-rich repeat kinase 2 (LRRK2), a protein associated with Parkinson′s disease. RAB32 is hypermethylated in microsatellite-unstable colon, gastric and endometrial adenocarcinomas.
Biochem/physiol Actions: Rab32 facilitates mitochondrial anchoring of protein kinaseA (PKA) and mitochondrial dynamics. Rab32 was also found to be required for the formation of autophagic vacuoles.

General description: The gene RAB32 (Ras-related protein) is mapped to human chromosome 6q24.3. It is a Rab GTPase which is present in the mitochondria. The protein also has a transient phagosomal localization and is also present at the melanosome membrane.
