Enhanced Validation-By Independent Antibodies Immunofluoresence. HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 2 μg/mL Anti-phospho-TDP-43 (pSer410) antibody produced in Rabbit (Cat. No. SAB4200224) (A). The antibody developed using Goat Anti-Rabbit IgG, FITC Conjugate antibody, and 2 μg/mL Anti-phospho-TDP-43 [pSer409] antibody produced in Rabbit (Cat. No. SAB4200223) (B). The antibody was developed using Goat Anti-Rabbit IgG, Cy3 Conjugate antibody. Results Two Anti phospho-TDP-43 antibodies, SAB4200224 (A) and SAB4200223 (B) target different regions of phospho-TDP-43 show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunoblotting Cells extracts were separated on SDS-PAGE, blotted with 1 μg/mL Anti-phospho-TDP-43 [pSer410] (Cat. No. SAB4200224), and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. General description: TDP-43 (TAR DNA binding protein) belongs to the family of heterogeneous nuclear ribonucleoproteins (hnRNPs), that binds single stranded RNA. TDP-43 has been implicated in the transcription regulation of HIV. TDP-43 is the major ubiquinated component of cytoplasmic inclusions in frontotemporal lobe degeneration subtype FTLD-U and amyotrophic lateral sclerosis (ALS). TDP-43 is predominantly localized to the nucleus.
Immunogen: synthetic peptide containing phosphorylated Ser410 of human TDP-43, conjugated to KLH. The corresponding sequence is identical in mouse TDP-43.
Physical form: Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
