Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-Tensin 1 antibody produced in Rabbit (Cat. No. SAB4200283) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing Tensin 1 (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1.5 μg/mL.
Immunohistochemistry Localization of tensin 1 at sarcomeric fibers in rat heart. Formalin-fixed paraffin-embedded tissue was stained with 10 μg/mL Anti-Tensin 1 (Cat. No. SAB4200283) followed by Goat Anti-Rabbit IgG, HRP conjugate (×20 magnification).
Immunofluorescence Cytoplasmic and focal adhesion localization of tensin 1 in chicken embryo fibroblast cells. Fixed cells were stained with 3 μg/mL Anti-Tensin 1 (Cat. No. SAB4200283) followed by Goat Anti-Rabbit IgG, Cy3 conjugate (×40 magnification). Nuclear staining with DAPI.
Immunoblotting Cell and tissue extracts were separated on SDS-PAGE, blotted with 1.5 μg/mL Anti-Tensin 1 (Cat. No. SAB4200283), and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. A10 2. Rat heart 3. HEK293-T overexpressing human tensin 1 4. HEK293-T non-transfected 5. A10: Antibody + immunizing tensin 1 peptide 6. HEK293-T overexpressing human tensin 1. Antibody + immunizing tensin 1 peptide

Biochem/physiol Actions: Tensin 1 (TNS1) contributes to transforming growth factor (TGF)-β-induced myofibroblast differentiation and extracellular fibronectin and collagen matrix formation. Disruption in the gene and associated signaling might hinder tissue fibrosis. The encoded protein anchors to stress fibers by interacting with cytoplasmic tails of integrins and increases metastatic abilities of cancer cells. Tensin plays a vital role in activation of the c-Jun N-terminal kinases (JNK) and p38 pathways.
Biochem/physiol Actions: Tensin 1 associates with actin, integrin, FAK, p130Cas, DLC-1 and protein phosphatase PP1 which affects cell polarization, migration and invasion. Tensin 1 undergoes extensive Ser/Thr and Tyr phosphorylation induced by extracellular matrix (ECM), various growth factors and oncogenes. In addition, tensin 1 is cleaved by calpain II, a focal adhesion protease, suggesting that tensin 1 plays important roles in organizing the actin cytoskeleton and mediating signal transduction.

General description: Tensin 1 (TNS1) is encoded by the gene mapped to human chromosome 2q35. The encoded protein is localized to focal adhesions and is mainly expressed in renal tubular epithelial cells (TECs).
Immunogen: synthetic peptide corresponding to a sequence located in the internal region of human tensin1, conjugated to KLH. The corresponding sequence is identical in rat and mouse tensin1.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
