Immunoblotting Lysate of HEK-293T cells co-transfected with murine CD3#ζ and constitutively active Lck (Lck-Y505F) was separated on SDS-PAGE and probed with Rabbit Anti-phospho-CD3#ζ (pTyr142) (Cat. No. SAB4200334). The antibody was developed with Goat anti-Rabbit IgG, peroxidase conjugate (Cat. No. A0545) and a chemiluminescence substrate. Lanes 1. Antibody: 0.5 MUg/mL 2. Antibody: 1 MUg/mL + 1MUg/mL non-phosphorylated form of the immunizing peptide 3. Antibody: 1 MUg/mL + 1 MUg/mL immunizing phosphopeptide.
Immunoprecipitation Rabbit Anti-phospho-CD3#ζ (pTyr142) (Cat. No. SAB4200334) was used to immunoprecipitate CD3#ζ from lysate of HEK-293T cells co-transfected with murine CD3#ζ and constitutively active Lck (Lck-Y505F). Detection antibody: mouse Anti-CD3#ζ followed by Anti-Mouse IgG (Fc specific)-Peroxidase (Cat. No. A2554). Lanes 1. Negative control: IP antibody without lysate 2. IP antibody: 4 MUg antibody 3. IP antibody: 2 MUg antibody 4. IP antibody: 1 MUg antibody

Application: Anti-phospho-CD3ζ (pTyr142) antibody produced in rabbit has been used in several immunochemical techniques including immunoblotting and immunoprecipitation.
Biochem/physiol Actions: Amongst the TCR subunits, the CD3 ζ chain plays a key role in regulating TCR assembly, transport, and cell surface expression. Down-regulation of TCR ζ chain expression associated with T cell dysfunction was described in various pathologies.

General description: The TCR-CD3 multimeric complex is composed of TCRαβ, CD3γε and CD3δε and TCR‐ζ homodimer. The CD3γ, CD3δ and CD3ε, each contain one immunoreceptor tyrosine-based activation motif (ITAM), whereas the ζ chain contains three ITAMs.
Immunogen: synthetic phosphopeptide corresponding to residues surrounding Tyr142 of mouse CD3ζ, conjugated to KLH. The corresponding sequence is identical in human CD3ζ.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
