Immunofluorescence HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton-X100. Fixed cells were stained with 1 μg/mL Rabbit Anti-Golph3 (N-terminal) (Cat. No. SAB4200341). The antibody was developed using Goat Anti-Rabbit IgG, Cy3 conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting Cell line lysates were separated on SDS-PAGE and probed with 2 μg/mL Rabbit Anti-Golph3 (N-terminal) (Cat. No. SAB4200341). The antibody was developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. HeLa; 2. WI-38; 3. NCI-H1299; 4. LA4; 5. NIH-3T3; 6. PC-12
Immunoprecipitation Rabbit Anti-Golph3 (N-terminal) (Cat. No. SAB4200341) was used to immunoprecipitate Golph3 from HeLa whole cell extract. Detection antibody: Rabbit Anti-Golph3 (C-terminal) (Cat. No. SAB4200340). Lanes 1. 5 μg antibody 2. 10 μg antibody 3. Negative control: 10 μg non-relevant antibody

Biochem/physiol Actions: Golph3 (Golgi phosphoprotein 3) is important for Golgi trafficking and morphology by interacting with the unconventional myosin MYO18A, linking Golgi membranes to the actin cytoskeleton. It is a phosphatidylinositol-4-phosphate (PtdIns(4)P binding protein required for Golgi function. GOLPH3 overexpression is correlated with hyperactivation of mTOR signaling, in human cells.

General description: Golph3 (Golgi phosphoprotein 3), also known as coat protein GPP34, is a peripheral membrane protein of the Golgi stack, that localizes to the trans-Golgi network. It is an oncogene that is commonly targeted for amplification in human cancer and in cancer cell lines.
Immunogen: peptide corresponding to the N-terminal region of human Golph3, conjugated to KLH. The corresponding sequence is identical in mouse, rat, monkey and bovine.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
