| Biochem/physiol Actions: | SMAD7 (mothers against decapentaplegic homolog 7) is a modulator of TGFβ (transforming growth factor β) signaling in immune cells which are associated with ulcerative colitis and inflammatory bowel syndrome. It plays a major role in the etiology of CRC (colorectal cancer). In addition, it also acts as a mediator of the negative feedback loop for both the TGFβ and BMP (bone morphogenetic proteins) signaling pathways. It is essential for the remodelling of pharyngeal artery and the enlargement of great vessel. In mouse, homozygous removal of Smad7 causes primarily fourth-related arch artery defects. Silencing of Smad7 in RCD (refractory coeliac disease) biopsy samples can decrease the expression of interleukin-6 and tumour necrosis factor-α. Polymorphism of this gene is associated with colorectal cancer. SMAD7 is an inhibitor of the TGFβ (transforming growth factor beta) /BMP (bone morphogenetic proteins) pathway. |
