Immunofluorescence Mouse NIH-3T3 cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton™ X-100. Fixed cells were stained with 5 μg/mL Rabbit Anti-SMAD6 (C-terminal) (Cat. No. SAB4200383). The antibody was developed using Goat Anti-Rabbit IgG, Cy3 conjugate.
Immunoblotting Whole extract of HEK-293T cells overexpressing human SMAD6 (C-terminal) was separated on SDS-PAGE and probed with Rabbit Anti-SMAD6 (C-terminal) (Cat. No. SAB4200383). The antibody was developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Biochem/physiol Actions: SMAD family member 6 (SMAD6) induces endocytosis and negatively regulates apoptosis by inhibiting transforming growth factor-β (TGF-β) signaling pathway. In addition, it also indirectly regulates stemness by inhibiting erythropoiesis in cord blood hematopoietic stem cells (HSCs). Mutation in the gene leads to congenital cardiovascular malformation (CVM). Overexpression or genetic variation of SMAD6 is associated with the development of ovarian cancer.

General description: SMAD family member 6 (SMAD6) is encoded by the gene mapped to human chromosome 15q22.31. The encoded protein is localized in both nuclei and cytoplasm and is expressed in variety of human tissues, including ovary. SMAD6 consists of MAD homology 2 (MH2) domain, involved in protein-protein interaction.
Immunogen: peptide corresponding to the C-terminal region of human SMAD6, conjugated to KLH. The corresponding sequence is identical in rat, mouse and bovine.
Physical form: Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
