Immunofluorescence A549 cells were fixed and permeabilized with cold methanol followed by acetone. Fixed cells were stained with 5 μg/mL Mouse Anti-USP8 Clone: US872 (Cat. No. SAB4200527). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting MCF7 cell extract was separated on SDS-PAGE and probed with Mouse Anti-USP8 Clone: US872 (Cat. No. SAB4200527). The antibody was developed using Goat Anti-Mouse IgG-Peroxidase (Cat. No. A2304) and a chemiluminescent substrate. Lanes 1. 0.5 μg/mL antibody 2. 0.5 μg/mL antibody + USP8 immunizing peptide (human, 696-710)

Flow Cytometry 1×106 MCF7 cells were fixed and permeabilized with 2% Paraformaldehyde and 0.1% Triton X-100. Fixed cells were labeled with 5 μg Mouse Anti-USP8 Clone: US872 (Cat. No. SAB4200527) followed by Rabbit Anti-Mouse IgG, FITC conjugate. Green histogram: negative control without first antibody. Purple histogram: antibody specific staining.

Biochem/physiol Actions: Ubiquitin specific protease 8 (USP8) was found to regulate ubiquitination of frizzled, a receptor for Wnt/Wingless. Deletion of USP8 in mice causes embryonic lethality. The defect was accompanied by a strong reduction or absence of several growth factor receptor tyrosine kinases (RTKs), like EGFR, hepatocyte growth factor receptor (c-met), and ERBB3.USP8 inactivation results in enhanced ubiquitination of ligand-activated EGFR. USP8 in conjunction with components of the ESCRT-0 complex, plays an integral role in the early endosomal sorting machinery that functions to protect epidermal growth factor receptor (EGFR) as well as other receptors from lysosomal degradation.

General description: Monoclonal Anti-USP8 (mouse IgG1 isotype) is derived from the hybridoma US872 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice. Ubiquitin specific protease 8 (USP8) belongs to the family of de-ubiquitinating enzymes (DUBs).
Immunogen: synthetic peptide corresponding to an internal sequence of human USP8, conjugated to KLH.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
