Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-Claudin-1 (C-terminal) antibody produced in Rabbit (Cat. No. SAB4200534) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing Claudin-1 (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 0.5 μg/mL.
Immunohistochemistry Localization of claudin-1 in human colon. Formalin-fixed paraffin-embedded tissue was stained with 20 μg/mL. Anti-Claudin-1 (C-terminal) (Cat. No. SAB4200534) followed by Biotin-Goat Anti-Rabbit IgG/ExtrAvidin®HRP (×20 magnification).
Immunofluorescence Membrane localization of claudin-1 in MDCK cells. Fixed cells were stained with 1 μg/mL Anti-Claudin-1 (C-terminal) (Cat. No. SAB4200534) followed by Goat Anti-Rabbit IgG, Cy3 conjugate (×40 magnification). Nuclear staining with DAPI.
Immunoblotting Cell lysates of HEK-293T overexpressing human claudin-1 (lanes 1,3) and HEK293-T non-transfected (lane 2), were separated on SDS-PAGE, blotted with Anti-Claudin-1 (C-terminal) (Cat. Biochem/physiol Actions: Claudin-1 directly interacts with tight junction (TJ) -associated peripheral membrane proteins zonula occludens (ZO) proteins -1/2/3. Defects in the gene encoding claudin-1 are the cause of an autosomal recessive syndrome named ichthyosis-sclerosing cholangitis neonatal (NISCH). Claudins′ expression is altered in several human cancers. Claudin-1 is frequently up-regulated in colorectal carcinomas (CRCs), resulting in tumor differentiation and progression.

General description: Claudin-1, also known as claudin 1 (CLDN1), seminal metalloprotease-1 (Semp1) and ichthyosis, leukocyte vacuoles, alopecia, and sclerosing cholangitis (ILVASC), is expressed at high levels in kidney and liver and at lower levels in spleen, heart, brain, lung and testis. Claudins are characterized with four transmembrane domains and two extracellular loops, required to form tight junction strands.
Immunogen: synthetic peptide corresponding to a sequence at the C-terminus of human claudin-1, conjugated to KLH. The corresponding sequence is highly conserved in mouse claudin-1 (single amino acid substitution) and in rat claudin-1 (90% identity).
Physical form: Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
