Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-Myocardin antibody produced in Rabbit (Cat. No. SAB4200539) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing Myocardin (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1 μg/mL
Immunohistochemistry Staining of formalin-fixed, paraffin-embedded Human heart sections with 10 μg/mL Rabbit Anti-Myocardin (Cat. No. SAB4200539) using biotin/ExtrAvidin®-Peroxidase.
Immunohistochemistry Staining of formalin-fixed, paraffin-embedded Chicken heart sections with 10 μg/mL Rabbit Anti-Myocardin (Cat. No. SAB4200539) using biotin/ExtrAvidin®-Peroxidase.
Immunofluorescence
A10 cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 2 μg/mL Rabbit Anti-Myocardin (Cat. No. SAB4200539). The antibody was developed using Goat Anti-Rabbit IgG, FITC conjugate. Cells were counterstained with phalloidin rhodamine conjugate to stain Actin (red).
Western Blotting Cell line lysates were separated on SDS-PAGE and probed with 1 μg/mL Rabbit Anti-Myocardin (Cat. No. SAB4200539). The antibody was developed using Goat Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. HCT-116 2. HEK-293—overexpressing Myocardin 3. SW-480 4. Untransfected HEK-293 5. HCT-116 + Myocardin immunizing peptide

Biochem/physiol Actions: Myocardin (MYOCD) acts as an important smooth and cardiac muscle-specific transcriptional coactivator of serum response factor (SRF). It also plays an essential role in smooth muscle cell (SMC) differentiation. The encoded protein regulates bone morphogenetic protein 10 (BMP10) expression and is required for cardiac development, as well as cardiomyocyte survival and maintenance of heart function. MYOCD expression is associated with the development of atherosclerosis, Alzheimer’s disease and malignant transformation.

General description: Anti-Myocardin specifically recognizes human, rat and chicken myocardin (MYOCD). Myocardin is encoded by the gene mapped to human chromosome 17p11.2. It contains a conserved SAP (SAF-A/B, Acinus, and PIAS) domain, involved in the regulation of nuclear organization, chromosomal dynamics and apoptosis. The encoded protein is expressed in a precise developmentally regulated, lineage-restricted pattern in the embryo and during postnatal development.
Immunogen: a synthetic peptide corresponding to an internal sequence of human myocardin, conjugated to KLH. The corresponding sequence is identical in human myocardin isoform 2 and in rat and mouse myocardin.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
