Immunohistochemistry Staining of formalin-fixed, paraffin-embedded and antigen retrieval sections of Rat cerebellum with 10 μg/mL Mouse Anti-PARVALBUMIN Clone: PARV-19 (Cat. No. SAB4200545), using biotin/ExtrAvidin®-Peroxidase (Cat. No. B0529).
Immunofluorescence BLO-11 cells were fixed and permeabilized with 4% Paraformaldehyde and 0.5% Triton™ X-100. Fixed cells were stained with 10 μg/mL Mouse Anti-PARVALBUMIN Clone: PARV-19 (Cat. No. SAB4200545). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting Mouse muscle extract was separated on SDS-PAGE and probed with Mouse Anti-Parvalbumin Clone: PARV-19 (Cat. No. SAB4200545). The antibody was developed using Goat Anti-Mouse IgG-Peroxidase (Cat. No. A2304) and a chemiluminescent substrate. Lanes 1. 1 μg/mL antibody 2. Negative Control

Biochem/physiol Actions: Ca2+ participates in cell motility, release of neurotransmitters, membrane permeability, excitation−contraction of muscles, cell secretory procedures, and transmission of nerve impulses. Intracellular Ca2+ exchangers are essential to modulate these processes efficiently. Parvalbumin (PV), a calcium-binding protein, can regulate the intracellular calcium dynamics in neurons. Hence it may have an impact on facilitation and depression at synapses. Interestingly, PV deficiency alters neuronal activity, a key mechanism leading to epileptic seizures or Parkinson.

General description: Monoclonal Anti-Parvalbumin (mouse IgG1 isotype) is derived from the hybridoma PARV-19 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with purified frog muscle parvalbumin. Parvalbumin (PV) is a member of the family of Ca2+-binding molecules. It carries three helix-loop-helix EF-hand Ca2+ binding motifs, with the two C-terminal motifs.
Immunogen: purified frog muscle parvalbumin. The isotype is determined by ELISA using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note: For extended storage, freeze at –20 °C in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
