Immunohistochemistry Staining of formalin-fixed, paraffin-embedded human duodenum sections with 20 μg/mL Rabbit Anti-APC (C-terminal) (Cat. No. SAB4200594) using biotin/ExtrAvidin®-Peroxidase.
Immunofluorescence Caco-2 cells were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 2 μg/mL Rabbit Anti-APC (C-terminal) (Cat. No. SAB4200594). The antibody was developed using Goat Anti-Rabbit IgG, Cy3 conjugate. Cells were counterstained with DAPI (blue) to stain nuclei. Exposure/vibrancy may have been enhanced for clarity.
Western Blotting Cell line lysates were separated on SDS-PAGE and probed with 2.5 μg/mL Rabbit Anti-APC (C-terminal) (Cat. No. SAB4200594). The antibody was developed using Goat Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1. WiDr 2. HT-29 3. HepG2 4. SW620 5. HEK-293 6. HeLa

Biochem/physiol Actions: Adenomatous Polyposis Coli (APC) has a key role in the proteasome-mediated degradation of β-catenin. APC mutations cause accumulation of β-catenin in the nucleus leading to activation of LEF-1 and/or TCF, and the induction of target genes such as the oncogene c-myc. It is involved in regulation of mitotic chromosome separation and stability.

General description: Adenomatous Polyposis Coli (APC) is a tumor suppressor gene. The APC gene encodes a large multifunctional protein that has critical functions in cell migration, adhesion and proliferation. It is a shuttling nucleo-cytoplasmic protein expressed in various tissues and its expression in the colorectal epithelium contributes to its normal growth and differentiation.
Immunogen: synthetic peptide corresponding to a sequence at the C-terminal region of human APC, conjugated to KLH. The corresponding sequence is identical in human APC isoform b and highly conserved (single amino acid substitution) in rat and mouse APC.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
