Immunofluorescence A375 cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 2.5 μg/mL Monoclonal Anti-NG2/CSPG4, Clone: 2164B6 (Cat. No. SAB4200621). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate.
Flow Cytometry 1×106 SK-Mel-28 cells were fixed and permeabilized with 2% Paraformaldehyde followed by 0.1% Triton X-100. Fixed cells were labeled with 2.5 μg/test Monoclonal Anti-NG2/CSPG4, Clone: 2164B6 (Cat. No. SAB4200621) followed by Rabbit Anti-Mouse IgG, FITC conjugate. Green histogram: negative control without first antibody, Purple histogram: antibody specific staining.

Biochem/physiol Actions: Nerve/glial antigen 2 (NG2) plays a central role in linking multiple oncogenic pathways required for malignant progression. NG2 expression in radial growth phase (RGP) human melanomas enables migration, protease activation and epithelial to mesenchymal transition, which is important in primary tumors for facilitating progression from a radial to vertical growth phase phenotype. It regulates sustained, high-level activation of key survival and growth pathways, integrin-regulated focal adhesion kinase (FAK), extracellular signal-regulated kinases 1 and 2 (ERK 1,2) and phosphoinositide 3-kinase (PI3K)/AKT pathways.
