Immunofluorescence HeLa cells were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 10 μg/mL Monoclonal Anti-ALIX/PDCD6IP, Clone: ALIX-1 (Cat. No. SAB4200646). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate.
Immunoblotting Whole extract of human MOLT4 cells was separated on SDS-PAGE and probed with Monoclonal Anti-ALIX/PDCD6IP, Clone: ALIX-1 (Cat. No. SAB4200646). The antibody was developed using Goat Anti-Mouse IgG-Peroxidase (Cat. No. A2304) and a chemiluminescent substrate. Lanes 1. 2 μg/mL antibody 2. 5 μg/mL antibody 3. Negative control: without first antibody 4. Positive control: Rabbit Anti ALIX (Cat. No. SAB4200476)
Immunoprecipitation Monoclonal Anti-ALIX/PDCD6IP, Clone: ALIX-1 (Cat. No. SAB4200646) was used to immunoprecipitate ALIX/PDCD6IP from Whole extract of human MOLT4. Detection antibody: Rabbit Anti-ALIX (C-terminal) (Cat. No. SAB4200477), followed by Anti-Rabbit IgG-peroxidase (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1.10 μg/test antibody 2. 20 μg/test antibody 3. Negative control: 10 μg/test non-relevant antibody 4. Whole extract of human MOLT4 cells.
Flow Cytometry 1×106 MOLT4 cells were fixed and permeabilized with 2% Paraformaldehyde followed by 0.1% Triton X-100. Fixed cells were labeled with 5 μg/test Monoclonal Anti-ALIX/PDCD6IP, Clone: ALIX-1 (Cat. No. SAB4200646) followed by Rabbit Anti-Mouse IgG, FITC conjugate. Green histogram: negative control without first antibody, Purple histogram: antibody specific staining.

Biochem/physiol Actions: ALIX (Apoptosis-linked gene 2 (ALG-2)-interacting protein X) plays a role in ESCRT (endosomal sorting complexes required for transport) pathway. It is involved in the abscission stage of cytokinesis, in intralumenal endosomal localization, and in enveloped virus budding. Overexpression of this protein can block apoptosis. In addition, ALIX binds to PDCD6, a protein required for apoptosis, in a calcium-dependent manner. ALIX also binds to endophilins, proteins that regulate membrane shape during endocytosis. Overexpression of ALIX and endophilins results in cytoplasmic vacuolization, which may be partly responsible for the protection against cell death.

General description: Anti-ALIX/PDCD6IP antibody, Mouse monoclonal (mouse IgG1 isotype) is derived from the hybridoma ALIX-1 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a peptide corresponding to an internal region of human ALIX/ PDCD6IP, conjugated to KLH. ALIX (Apoptosis-linked gene 2 (ALG-2)-interacting protein X), also named Programmed Cell Death 6 interacting protein (PDCD6IP).
