| Biochem/physiol Actions: | Signal transducer and activator of transcription 3 (STAT3) plays a vital role in various cellular responses such as induction of an acute-phase response in hepatoma cells, stimulation of proliferation in B lymphocytes, activation of terminal differentiation and growth arrest in monocytes, and regulation of the pluripotency of embryonic stem cells. Activated STAT3 has been involved in malignant transformation. STAT3 may be a key player in the pathogenesis of diverse human cancers which makes this molecule a prime target for novel therapies. Phospho-tyrosine residue 705 (Tyr705) Stat3 can be used as a potential marker for determining overall survival of node-negative breast cancer patients. Phosphorylated STAT3 might be involved in angiogenesis, anti-apoptosis and tumor progression. |

The gene encoding protein consists of 24 exons and is extended over a length of 75170 bases on genomic DNA STAT3 is mapped on human chromosome 17q21.2. The protein is characterized with a conserved amino-terminal domain, a DNA-binding domain, an SH2 domain and a carboxy-terminal transactivation domain. |
