| Biochem/physiol Actions: | Protein phosphatase 2 regulatory subunit B alpha (PPP2R2A) protein complexes are critical for many cellular processes including cell growth and differentiation, cell motility, DNA damage response, progression of cell cycle and apoptosis. PP2A/B55 dephosphorylates and activates heat shock factor 1 (HSF 1) directly for various cellular processes. Family with sequence similarity 122A (FAM122A) is a endogenous inhibitor of phosphatase activity of PP2A/B55. Overexpression or dysregulation of PP2A has several implications in the developmental stage, embryonic and neonatal death. Polymorphism of PP2A leading to dysregulation is associated with Alzheimer′s disease and various malignancies. Mutations leading to enhanced activity causes asthma, type II diabetes, chronic obstructive lung disorder. Suppression of PPP2R2A expression by micro-RNA892a (mi-R 892a) promotes the growth of colorectal cancer cells. Somatic deletion mutation of PPP2R2A have a potential role in prostate cancer. PPP2R2A functions as a tumour suppressor gene in numerous cancer types. Downregulation of PPP2R2A along with upregulation of cyclin D1 causes aggressiveness in luminal-like breast cancer. PPP2R2A is known to dephosphorylate Tau protein, whose hyperphosphorylation is associated with Alzheimer′s′ disease. |

| General description: | Protein phosphatase 2 regulatory subunit B alpha (PPP2R2A or PP2A/B55) is a heterotrimeric holoenzyme consisting of a scaffolding subunit (A), a regulatory subunit (B) and a catalytic subunit (C). The B subunit, based on the homology is classified into four families, and each has several isoforms. B55 subunit is ubiquitously expressed among all the families of PP2A. In human chromosome, the gene PPP2R2A is localized on 8p21.2. |
