| Biochem/physiol Actions: | Peroxisome proliferator-activated receptor γ (PPARγ) induces the expression of genes coding for proteins implicated in glucose and lipid metabolism. Natural PPAR-γ agonists found in foods functions as an anti-inflammatory molecule, thus, PPARγ is not only a target of the pharmaceutical industry, but also a novel target of the food industry. PPAR-γ regulates energy storage, adipocyte differentiation, and cellular inflammatory and ischemic responses. PPARγ is also involved in the production of prostanoids that regulate vascular function. PPARγ agonists, thiazolidinediones including rosiglitazone and pioglitazone, enhance insulin sensitivity by inhibiting the tumor necrosis factor α (TNF-α) activity in adipocytes. These PPAR-γ agonists are used in the treatment of type 2 diabetes. PPARγ functions as a potential therapeutic target in pulmonary hypertension. |

| General description: | Peroxisome proliferator-activated receptor γ (PPARγ) is one of the subtypes of PPAR, and belongs to the nuclear hormone receptor superfamily. PPARγ is further sub classified into four isoforms and they have tissue specific expression. PPARγ is encoded by the gene mapped to human chromosome 3p25. PPARγ contains N- terminal AF (activation function)-1 transactivation domain, followed by a DNA-binding domain and a dimerization and ligand-binding domain. |
