Biochem/physiol Actions: | Cluster of differentiation 43 (CD43) is known to regulate cell cycle and cell relocation in both lymphoid and non-lymphoid tumor cells. It serves as a biomarker in chronic lymphocytic leukemia and other hematologic cancer diagnosis. CD43 is associated with cell movement, cell differentiation, apoptosis and immune homeostasis. Glycosylation and sialylation of CD43 at extracellular C-terminus, enables its purpose as a ligand for selectins. CD43 promotes extracellular signal-regulated kinase 1/2 (ERK 1/2) signaling, which is necessary for actin cytoskeleton modifications. CD43 is associated with autoimmune diseases, including systemic lupus erythematosus, diabetes, human immunodeficiency virus (HIV) infection and Wiskott-Aldrich syndrome. |

General description: | CD43 is one of the major glycoproteins of thymocytes and T lymphocytes. It plays a role in the physicochemical properties of the T cell surface and in lectin binding. CD43 presents carbohydrate ligands to selectins. It has an extended rodlike structure that could protrude above the glycocalyx of the cell and allow multiple glycan chains to be accessible for binding. The antigen is a counter receptor for SN/Siglec1. During T cell activation CD43 is actively removed from the T cell antigen presenting cell contact site suggesting a negative regulatory role in adaptive immune response. |
General description: | The SPN (sialophorin) gene is mapped to human chromosome 16p11.2 and encodesa transmembrane sialoglycoprotein cluster of differentiation 43 (CD43). It is localized to the surface of leukocytes and is expressed in hematopoietic cells. CD43 protein structure possesses a 23-amino acid transmembrane bridge connecting the N- and C- terminal regions. Its cytoplasmic tail is highly conserved. |
