Immunocytochemistry Immunocytochemical staining of HeLa cells using Anti-Histone H3 (Unmodified Lys4) (Clone RM186) (red). Actin filaments have been labeled with fluorescein phalloidin (green), and nuclei stained with DAPI (blue).
Immunoblotting Western Blot of acid extracts of HeLa cells treated with sodium butyrate (HeLa+NaBu), treated with Nocodazole (HeLa+Noc), or non-treated; and recombinant Histone H3.1 protein, using RM186 at 1 mg/mL, showed a band of Histone H3 unmodified at Lys4.
Chart RM186 specifically recognizes Histone H3 unmodified at Lys4 and does not recognize acetylated, monomethylated, dimethylated, or trimethylated Lys4. The antibody binding specificity allows for modifications of Arg2 or Thr3 in Histone H3.
