Western Blotting Western blot analysis of extracts of various cell lines, using Phospho-mTOR-S2481 (SAB5701822) at 1:1000 dilution.Both 293T cells and NIH/3T3 cells were treated by IGF-1 (50 ng/mL) at 37 °C for 5 minutes after serum-starvation overnight. Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) at 1:10000 dilution. Lysates/proteins: 25 μg per lane. Blocking buffer: 3% BSA. Detection: ECL™ Basic Kit (GERPN3004). Exposure time: 3 min.
