Activity Assay The activity of recombinant human Antithrombin III (Cat. No. SAE0102) is measured by its ability to inhibit thrombin’s activity in the presence of heparin. The IC50 is defined as the effective concentration of AT-III that inhibits 50% of thrombin’s activity.

Application: This recombinant human AT-III product can be used to study the mode of action of the enzyme, as well as its potential inhibitors. It may also be used as a standard, e.g. in antithrombin activity assay, and/or in in vivo studies.
Biochem/physiol Actions: Antithrombin III (AT-III), or Serpin C1 is a serine protease inhibitor glycoprotein, is a member of the Serpin superfamily. Antithrombin III inactivates several components in the coagulation system, IIa,IXa and mainly thrombin and factor Xa. The inhibitory activity of Antithrombin III is potentiated and increased by the polysaccharide heparin. In addition to its abolishment of coagulation, Antithrombin III was also found to suppress proinflammatory mediators, in part by its anticoagulation action. Congenital antithrombin deficiency results in loss of Antithrombin III activity, and leads to thrombosis. Thus, human Antithrombin III enzyme may be prescribed for the treatment and prevention of thromboembolism, as well as for the prevention of perioperative and peripartum thromboembolism.
General description: Recombinant human antithrombin III (AT-III) is expressed in human HEK 293 cells as a glycoprotein with a calculated molecular mass of 49 kDa (amino acids 33-464). This protein is manufactured in human cells, with no serum. The human cells expression system allows human-like glycosylation and folding, and often supports higher specific activity of the protein. The protein is produced with no artificial tags.
