| Biochem/physiol Actions: | BTC is a selective estrogen receptor (ER) modulator (SERM) that acts as a selective estrogen mimic (SEM). BTC has been found to be both estrogenic and antiproliferative in ER-positive endocrine-resistant breast cancer cell lines that overexpress protein kinase C alpha (PKCα), which is often associated with tamoxifen resistance. BTC caused regression of endocrine-resistant breast cancer xenografts, and did not support the growth of hormone-dependent xenografts in vivo or cause an increase in uterine weight. As in the case with raloxifene and estradiol, BTC tumor regression was accompanied by the exit of ERα from the nucleus, suggesting extranuclear ERα may be involved in the mehanism of action. |
