β-Glucuronidase is routinely used to hydrolyze glucuronide-conjugates of drug metabolites in order to optimize sample preparation prior to LC-MS, GC and ELISA analysis.
LC-MS analysis of codeine-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. The limpet-derived enzyme G8132 performed best with this substrate, with reaction completion within 3–4 hours at 60 °C at 1 unit per mL. Comparative data above was performed at 60 °C at 10 unit per mL.
LC-MS analysis of hydromorphone-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. Molluscan enzymes performed similarly. The reaction was complete within 3–4 hours at 60 °C at 1 unit per mL. The E. coli enzymes exhibited low specificity for this substrate.

Biochem/physiol Actions: Beta-glucuronidase is anenzyme present in several animals, plants, and bacteria. It catalyzes thehydrolysis of Beta-glucuronide conjugates and plant glucuronides.Beta-glucuronidase is a member of the glycosyl hydrolases (GHs) family 2 and is ahomo-tetrameric glycoprotein. It facilitates the removal of glucuronic acidsfrom glucuronides and aids in the liver metabolism enzymes for the excretion ofendo- and xeno-biotics.
Other Notes: One Sigma or modified Fishman unit will liberate 1.0 μg of phenolphthalein from phenolphthalein glucuronide per hr at 37 °C at pH 3.8 (30 min assay).
