Biochem/physiol Actions: | Nuclear receptors form the largest known family of transcription factors and have a crucial role in nearly all aspects of vertebrate development and adult physiology by transducing the effects of hormones into transcriptional responses. The family is defined by two domains: (a) the central, highly conserved, DNA-binding domain (DBD) of approx. 66 amino acids, and (b) the C-terminal, structurally conserved, ligand-binding domain (LBD) of approx. 250 amino acids. The amino-terminal regions are least conserved among nuclear receptor sequences. Retinoids, the natural and syntheic derivatives of vitamin A, have been shown to inhibit the growth of many human tumor cells. Although the exact mechanism for this growth suppression remains unknown, the importance of retinoic acid receptors (RARs) and retinoid X receptors (RXRs) have been established in a number of tumor cell models. Three types of RARs have been identified (RAR, RAR, and RAR ) with homologous but slightly different retinoic acid (RA) binding domains. Transactivation assays and retinoid binding assays with RAR, RAR, and RAR have shown that each RAR has a different affinity for RA and RA analogs. |
