Biochem/physiol Actions: | CDK5 is a neuron-specific kinase that plays a vital role in the development, functioning, and diseases of the central nervous system. Its optimum functioning is highly essential for proper neuronal migration, differentiation, axonal elongation, and synaptic function. CDK5 modulates neurite outgrowth, and axonal growth by interacting with several substrates like N- cadherin, Rap Guanine Nucleotide Exchange Factor 2(RapGEF2), focal adhesion kinase (FAK), p21 activating kinase 1, serotonin 6 receptor and Ras-related protein 8. CDK5 also plays a key role in the formation of the synapse, its maintenance, and communication. It is involved in the formation and retraction of the dendritic spines. It also regulates neuron secretion at the synapse by phosphorylating key mediators such as Synaspsin1. In addition, CDK5, present in the synaptic membrane phosphorylates the erythroblastic leukemia viral oncogene homolog B receptor (ErbB), resulting in increased acetylcholine receptor transcription. Lastly, CDK5 is also involved in the regulation of various other cellular processes like cell cycle, cell survival, apoptosis, gene regulation, etc. |
