Biochem/physiol Actions: Matrix metalloproteinase-2 (MMP-2) cleaves the collagen-like sequence Pro-Gln-Gly-|-Ile-Ala-Gly-Gln. It is involved in diverse functions such as remodeling of the vasculature, angiogenesis, tissue repair, tumor invasion, inflammation, and atherosclerotic plaque rupture. It degrades type IV, V, VII, and X collagens. The protein degrades extracellular matrix proteins and acts on several non-matrix proteins such as big endothelial 1 (bET) and β-type calcitonin gene-related peptide (CGRP) promoting vasoconstriction. MMP-2 also cleaves KISS (metastasis suppressor) at a Gly-|-Leu bond. The protein appears to have a role in myocardial cell death pathways and contributes to myocardial oxidative stress by regulating the activity of glycogen synthase kinase-3 β (GSK3β). It has been shown to cleave GSK3β in vitro. PEX, the C-terminal non-catalytic fragment of MMP2, possesses anti-angiogenic and anti-tumor properties and inhibits cell migration and cell adhesion to fibroblast growth factor-2 (FGF2) and vitronectin.
General description: Matrix metalloproteinase-2 (MMP-2) which is also known as 72kDa type IV collagenase, 72kDa gelatinase, gelatinase A and CLG4A, belongs to the peptidase M10A family. It contains three fibronectin type-II domains and four hemopexin-like domains. MMP-2 is produced by normal skin fibroblasts. The gene encoding this protein is localized on human chromosome 16q12.2.
Physical form: Lyophilized from 0.22 μm filtered solution in 50 mM Tris, 150 mM NaCl, pH 8.0. Generally 5-8% Mannitol or trehalose is added as a protectant before lyophilization.
Reconstitution: Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
