Immunohistochemistry Jagged-1 (Human): FC (Human) protein: Cat. No. SRP8012: Adipogenesis inhibition of 3T3L-1 cells. 3T3L1 cells were maintained in DMEM supplemented with 10% FBS and penicillin-streptomycin. When the cells were confluently grown adipogenesis was initiated by adding IBMX, Dexamethasone, and insulin to 0.5 mM, 1βM, and 10 βg/mL, respectively and continued for 2 days. The medium was replaced every 2 days with new medium containing insulin in the presence or absence of 5 βg/mL of Human Jagged-1 (Human): FC (Human) and Human CD137-Fc as a control-Fc. Staining with Oil Red O was typically performed on day 7.
Immunoblotting Jagged-1 (Human): FC (Human) protein: Cat. No. SRP8012: Induction of Hes-1 with the treatment of Jagged-1 (Human): FC (Human). A Mouse preadpipocyte cell line, 3T3L1, was stimulated with 5 βg/mL of Human Jagged 1-Fc as in indicated time points and each cell lysate was prepared and subjected to western blot by using Anti-Human Hes1 or GAPDH.

Application: Jagged-1 has been used to activate Notch1 signaling pathway that modulates cardiac AMPK (5′ AMP-activated protein kinase) signaling cascade. Jagged-1 (human): FC (human) has been used to determine the cardiac substrate (fatty acid and glucose) metabolism in working heart model.
Biochem/physiol Actions: The encoded protein is a transmembrane-type ligand for Notch family receptor and is capable of inducing Notch signaling, which participates in embryogenesis, tissue regeneration, and carcinogenesis. Jagged-1 is a WNT-dependent Notch signaling activator that is involved in the maintenance of homeostasis of stem and progenitor cells. Jagged proteins are found to be involved in development and carcinogenesis. Overexpression of Jagged-1 has been observed in cervical and breast cancers.
General description: Jagged-1 is a ligand for multiple Notch receptors and involved in the mediation of Notch signaling. It is involved in cell-fate decisions during hematopoiesis. Is involved in early and late stages of mammalian cardiovascular development. Inhibits myoblast differentiation. Enhances fibroblast growth factor-induced angiogenesis (in vitro). Defects in Jagged-1 are the cause of Alagille syndrome type 1 (ALGS1) and tetralogy of Fallot (TOF).
General description: Jagged-1, encoded by the gene JAG1, is a Serrate homolog found in vertebrates. It is expressed by both primitive and mature human hematopoietic cells. The encoded protein contains EGF (epidermal growth factor) repeats, transmembrane domain and a cysteine-rich region in between the two.
Other Notes: Signal peptide and extracellular domain of human Jagged-1 (HJ1; CD339) (aa 1-1067) are fused at the C-terminus to the Fc portion of human IgG1.
